Abstract

Objectives: To investigate the inhibitory effect of eplerenone, an aldosterone receptor blocker, on cell pyroptosis in contralateral kidney of UUO rats, and to elucidate the role of aldosterone in the process of unilateral renal injury-induced chronic renal failure. Methods: 30 male Wistar rats weight 190 ± 10 g after 7 days to adapt, rats are randomly divided into 3 groups: sham operation group, UUO group and eplerenone group. The rats in UUO and Eplerenone group had the UUO operation and treated with Eplerenone of 100 mg/Kg/d. After 10 days, the kidneys were removed, HE and Masson were used to detect pathological changes of kidney. The cell pyroptosis was detected by TUNEL method. The expressions of NLRP3, Caspase1, IL-1β and SGK-1 were detected by immunohistochemistry, Western blot, immunofluorescence and laser confocal microscopy. Results: Renal pathology showed that inflammatory cells was significantly increased in UUO rats, and the number of pyroptosis cells increased in renal tubular epithelial cells significantly. The expressions of SGK-1, NLRP3, Caspase1 and IL-1β were up-regulated with UUO and down-regulated by Eplernone. Pyroptosis cells also inhibited with it. Conclusion: Cell pyroptosis was induce in contralateral kidney of UUO rats because of mineralocorticoid receptor activation, Eplerenone has a protective effect by blocking the aldosterone.
