Abstract

Ischemic stroke is the main type of stroke, causing a large number of patients dead and disabled every year. Tribuloside (TBS) is a kind of natural saponin, which can improve cerebral blood flow and protect brain tissue. In the present study, we have successfully developed gold nanoparticle-modified TBS, which increase the aqueous solubility of TBS and named the SERS tags. Based on the label surface enhanced Raman scattering (SERS) technique, it enables the release of TBS from the nanoparticles’ surface induced by GSH can be monitored and realize real-time detection. SERS technique provided a nanomolar range release of TBS from the gold nanoparticle's surfaces after the injection of external GSH. In vivo SERS spectra of TBS was obtained from the brain of living mice after GSH treatment. The results showed that the nano-tribuloside could effectively reduce apoptosis and brain tissue damage in the neuronal cell ischemia-hypoxia injury model and the mouse focal cerebral infarction model induced by photochemical method. This study provides a new insight for ischemia treat, and provides a theoretical and experimental basis for clinical practice.
