Abstract

A sensitive and easily reproducible colorimetric assay has been developed for shikimic acid based on periodate oxidation in concentrated phosphoric acid. The periodate oxidation product reacts with thiobarbituric acid to produce a stable chromogen which, when extracted into cyclohexanone, has an absorption maximum at 535 mμ and an apparent extinction coefficient of 33000 liter M −1 cm −1 with respect to shikimic acid. This chromogen is similar to the thiobarbituric acid chromogen formed after periodate oxidation of deoxyribose. The colorimetric method is not specific for shikimic acid since deoxyribose derivatives as well as other shikimic acid related compounds yield similar thiobarbituric acid chromogens.
