Abstract

Pyoverdine is the primary siderophore produced byPseudomonas aeruginosa. Ferripyoverdine was prepared from the spent medium of iron-starved cells. To study the utilization of ferripyoverdine byP. aeruginosa, iron-starved cells were incubated in a medium containing57ferripyoverdine for 20, 40, 60, 120 and 360 minutes. Mossbauer spectra were measured of whole cell, outer membrane, inner membrane and soluble fraction samples prepared from aliquots removed at these times. While no ferripyoverdine was detected in any sample, other ferric and/or ferrous species were detectable, leading to the conclusion that ferripyoverdine does not accumulate in the cell. Inner membranes and soluble fraction samples removed at 120 and 360 minutes show two ferric species as compared with one ferric species seen in samples removed at 20, 40, and 60 minutes.
