Abstract

Available protocols for stripping antibodies from immunoblots involve the use of sodium dodecyl sulfate (SDS) or low-pH buffers. SDS was shown to remove transferred proteins from membranes, and low-pH buffer was shown to inefficiently strip off antibodies. A solution containing 6 M guanidine hydrochloride, 0.2% nondenaturing detergent, and a reducing agent can rapidly strip off tightly bound antibodies from aged polyvinylidene fluoride (PVDF) immunoblots at room temperature without removing significant amounts of transferred protein.
