Abstract

A method is described for assessing the effect of putative stimulators and inhibitors of insect haemocyte activation in vitro. Monolayers of haemocytes were incubated with a known activator of the intracellular prophenoloxidase (proPO) pathway, the β1,3-glucan laminarin, and the production of active PO was visualised by the addition to the medium of DOPA, which was converted by PO to the black pigment, melanin. The number of black-staining cells were counted, and thus a direct numerical comparison of the effect of soluble molecules on proPO activation and of the effect of inhibitors on laminarin-induced stimulation, could be made. The method has been used to test for stimulatory and inhibitory products of the pathogenic fungus, Metarhizium anisopliae.
