Abstract
栃木県産マツタケの子実体から分離した菌株(F275)をHY培地へ移植し, 23℃で暗培養した. F275の培養菌糸をオオムギ培地へ移植して, 40～50日後に種菌を作製することができた. これがマツタケ菌であることはITS領域のシーケンスおよびマツタケ特異的プライマーを用いたPCRによって確認した. 試験管内に調製した素ジェランガム培地で発芽したアカマツ実生に4～5粒のオオムギ種菌を接種し, 培養9ヶ月後のアカマツ実生の側根を微分干渉顕微鏡で観察した結果, マツタケ菌が根の細胞間隙に侵入して菌根を合成した.
