Abstract

A rapid histological assay for the detection of virus-like particles (VLPs) within cells of Cylindrocapsa is reported. Cells containing abundant VLPs can be readily distinguished in this assay with the light microscope following staining with Schiff's reagent. Histochemical staining of the VLP aggregates suggests the particles contain DNA. The reliability of the assay was determined by selecting individual cells which gave a positive test for the presence of VLPs in the assay, and then isolating and processing the same cells for electron microscopy. Ultrastructural examination of such cells clearly confirmed the presence of VLPs. The application of this assay to screen other algae which may contain similar VLPs is discussed.
