Using computational and biological approaches, Sumida et al. show that IFNβ regulates expression of these receptors; expression of PD-1, LAG-3, and TIM-3 is induced, whereas expression of TIGIT is downregulated. These changes are elicited by a dynamic network of gene regulators that includes two mutually antagonistic modules of regulators with opposite temporal kinetics. The identification of new regulators of T-cell coinhibitory receptor expression has the potential to lead to new approaches to enhance ICB therapy for cancer. Sumida TS, …, Hafler DA.
