Abstract

Procedures for the demonstration of gram positive and negative microorganisms are a critical function of every histology laboratory but may yield inconsistent results. In addition, concerns about the safety of some components have recently been raised. This report describes a modification of the gram stain that eliminates the need for picric acid, acetone, ethyl ether and ethylene glycol monoethyl ether and makes use of a light green background. These modifications have not only improved the procedure's safety but have improved diagnostic interpretation by pathologists. This modification is highly reproducible and provides a rapid diagnosis of gram positive and negative bacterial infection. (The J Histotechnol 21:155, 1998)
