Abstract

The paper is intended to review on the available literature that are widely available regarding rolling circle amplification using improved Φ 29 DNA polymerase, an enzyme with unique property which make the amplification possible. Rolling circle amplification is in constant demand in order to replace present technologies such as PCR which require numerous reactants and much labour. This is why techniques with high sensitivity and performance which can reduce these limitations in DNA amplification and detection are very much important to the researchers. An extensive review of literature which describes the application of RCA in signal amplification to the sensitive detection of nucleic acids on microarrays demonstrated the flexibility of RCA as a universal detection/amplification module for integration with a variety of microarray substrates and nucleic acid detection formats. It also showed the ability of RCA to provide a large increase in sensitivity when employed as a signal amplifier in combination with contemporary methods for nucleic acid detection and allele discrimination. An overview on the application of rolling circle amplification in immunoassay(immunoRCA) as well as Rapid and Sensitive Detection of Severe Acute Respiratory Syndrome Coronavirus by Rolling Circle Amplification, are all provided by this paper.
