Abstract

We isolated and characterized from a Streptomyces species a new class-II restriction endonuclease, which recognizes the palindromic heptanucleotide sequence 5′-G G↓G W C C C 3′-C C C W G↑G G (where W = A or T) and cleaves double-stranded DNA after the second G in this sequence This Sse18251 enzyme cleaves phage λ DNA at one site, adenovirus type 2 DNA at eight sites, but does not cleave pBR322, SV40, CoIE1, pUC18 and pUC19, and replicative forms of M13mpl8 and M13mpl9, and ΦX174 DNAs.
