Abstract

A novel monoclonal antibody was raised against a syntheticN-myristoyl glycine that is characteristic of allN-myristoylated proteins. The immunoreaction suppressed in the presence of hemocyanin as well as albumin conjugated withN-myristoyl glycine and otherN-myristoyl glycyl peptides, while underivatized and myristoyl amino acid proteins or various fatty acids other than myristic acid exerted no effect. The antibody specifically reacted withN-myristoylated pp60c-srcin human colon adenocarcinoma cells,N-myristoylated pp60v-srcin Rous sarcoma virus-infected cells, andN-myristoylated Gag precursor protein Pr55gagin HIV-1-producing cells. Furthermore, the antibody immunoreacted with a newN-myristoylated p28gagderived from HIV-1gagprotein. The antibody is shown to be a very useful tool for identification ofN-myristoylated proteins.
