Abstract

Introduction: Survivin is expressed exclusively in cancer cells, which is considered to be a biological indicator of the chemo-resistance of several kinds of human carcinomas. In this study, we constructed an oncolytic HSV-1 vector (d120.survE) using survivin promoter which replicates selectively in survivin expressing cells. Then, we evaluated the ability of d120.survE and/or gemcitabine to inhibit the growth of pancreatic cancer cells. Methods: (1) Survivin promoter activity was measured using luciferase assay and survivin mRNA expression in cancer cells was quantified using TaqMAN PCR in the presence or absence of gemcitabine.
