Abstract

A new acidic amino acid (C 8H 15O 4NS) containing cysteine residue was isolated from the urine of an atherosclerotic patient. It melted at 213 ° with decomposition. Crystalline form is rhombic plate from 50% alcohol. Ninhydrin color is blue on paper chromatogram. R f values are 0.58-0.65 in butanol-acetic acid-water and 0.43-0.53 in phenol-water. The isolation method consists of desalting the urine by ion exchangers, preparing acidic amino acid fraction by Amberlite CG-45, and fractionating by cellulose column. The new amino acid was also found in the urine of cat and of hypothyroid patients such as those having myxoedema and cretinism. Another S-containing amino acid was occasionally found in those urine samples above described and was identical on two-dimensional paper chromatogram with a synthetic sample S-(2-methyl-2-carboxyethyl)cysteine.
