Abstract

A new modified noncarcinogenic histochemical staining technique for the localization of myeloperoxidase in peripheral blood and bone marrow cells using 4-chloro-1-naphthol as the chromogen and May-Grunwald-Giemsa (MGG) counterstain is described. Fifty specimens from healthy control subjects and patients with leukemia and refractory anemia with excess blasts in transformation were studied using the rapid myeloperoxidase staining technique as described by Elias and a modified approach using the MGG counterstain. We observed refined nuclear details in the latter group. This additional morphological information along with the demonstration of peroxidase activity offer superior identification of the leukemic blast cell population, which is critical to diagnosis and follow-up evaluation.
