Abstract

A new method for determining lipase activity in locust fat body is described. The procedure uses a micellar solution of triolein as substrate and demonstrates high and reproducible lipase activity in the fat body. The maximal activity is about 25-fold greater than that determined by using other substrates which have widely been used by many investigators. The activity depends on ionic strength with maximal activity observed at an ionic strength of 0.3. The optimal pH was 6.9. The V max was calculated to be about 2.6 μmol fatty acid/hr per g fat body and the apparent K m value was about 56 μM for triolein.
