Abstract

Characterization of Ewing’s sarcoma (ES) and primitive neuroectodermal tumor (PNET)-derived cells may be achieved by combined analysis of studies of secretion of specific cellular products, surface antigen analysis by monoclonal antibodies, and cytogenetic investigation. We have developed a new monoclonal antibody HBA-7/1, which was produced by immunization with a PNET-derived cell line ESIM-1. The HBA-7/1 antibody exhibits unique cell membrane reactivity with ES and PNET-derived cell lines and no reactivity with neuroblastoma or other malignant small round cell tumors. The epitope recognized by HBA-7/1 is preserved in paraffin-embedded tumor material in a highly reactive form. HBA-7/1 is directed to an as yet uncharacterized protein determinant expressed exclusively on cells of PNET orgin. Our results show common traits of neuroectoderm and ES-derived cells. Further study of HBA-7/1 and previously described antigens my contribute to the histological diagnosis of malignant small round cell tumors.
