Abstract

A method for quantitative estimation of Phytophthora cinnamomi Rands zoospores on plant roots is presented. 14C-Labeled zoospores are prepared from sporangia produced on mycelia grown in a medium supplemented with [U- 14C]glucose. Approximately 30% of the initial counts fed are incorporated into hyphae and 0.1% of these counts are incorporated into zoospores. There is a linear response between the number of counts in a preparation and spore numbers. 14C Counts in extracts of root tissue infected with 14C-labeled zoospores give a reliable estimate of zoospore numbers. The extent of zoospore adhesion to roots measured by 14C labeling and that measured by direct counting are comparable.
