Abstract

Analytical technologies with simple workflows and quick responses are urgently needed. Here, we report a homogeneous microchannel resistance sensor (hoMCRS) without washing steps or signal amplification. The specific recognition of target by biomolecules anchored on polystyrene (PS) microspheres induces aggregation of these insulating particles in solution, which can be statistically characterized using a homemade microchannel resistance detection system, thus to enable target quantification. The concept of hoMCRS is validated by immunoassay of procalcitonin and DNA hybridization assay of Salmonella enterica in real samples. This assay is performed without separation or washing steps, which simplifies the operation and reduces the cumulative error. In addition, the signal generation is free of enzymatic reaction or amplification, leading to a quick response and high stability. These merits make the hoMCRS appropriate for rapid and simple analyses in various settings with minimal operations and infrastructures.
