Abstract
A high affinity agonist . beta-adrenergic receptor complex is an intermediate for catecholamine stimulation of adenylate cyclase in turkey and frog erythrocyte membranes.
Highlights
Radioligand competition binding studies were performed usingthe antagonist [3H]dihydroalprenolol, and the data were analyzed by nonlinear least squares computer modeling procedures
It hasbeen reported that thebinding of agonists to turkey erythrocyteP-adrenergic receptors is to a single population of receptors of uniform affinity whichis not modulated by guanine nucleotides (9, 10)
Laboratory have directly demonstrated that agonists bind to Radioligand- bindingAssays-Frog erythrocyte membranes (0.2 the P-adrenergic receptor in frog erythrocyte membraneswith to 0.4 mg of protein/ml) or turkey erythrocyte membranes(0.4 to 0.6 mg of protein/ml) were incubated in a final volume of 0.5 to 1.0 ml with [“HIDHA (3to 5 nM) for 10min at 37°C
Summary
For each competition binding curve of [“HIDHA by competing agonist or antagonist, fitted parameter estimates were provided for the data, assuming either one or two affinity states of the receptor, with deviation of the observed pointsfrom the predicted values being weightedaccording to the preparations since Gpp(NH)p alone stimulates adenylatecyclase very little. Plex with the /I-adrenergic receptor which is not reversed by extensive washing or subsequent addition of (+)propranolol
