Histone variants contribute to epigenetic regulation in development and disease but require the chaperone machinery for correct deposition. In this issue of Molecular Cell, Ji etal.1 explain how the chaperone complex FACT removes the histone variant macroH2A1.2 and demonstrate its importance for gene activation in innate immune cells.
