Abstract

This chapter discusses a computerized biosensor system with on-line calibration for continuous ex vivo measurements of glucose and lactate. A biosensor system for continuous operation with on-line calibration, based on the enzymatic conversion of d-glucose or l-lactate has been developed. Sampling is performed continuously by means of a coaxial catheter and the sample stream is dialyzed on-line before passing through an enzyme reactor containing glucose oxidase or lactate oxidase immobilized to porous glass. The biosensor system has been used for measurements of glucose and lactate in bovine blood samples and the results compared with off-line analyses. The system is intended for continuous ex vivo measurements on human blood in medical applications requiring high operational stability. The calibration solution is injected at the tip of the sampling probe and thus the calibration procedure takes care of all sample handling steps as well as the analytical process itself. The sensor can be calibrated on-line without disconnecting it from the object or process monitored.
