Abstract
A technique for demonstration of glycosaminoglycans and other biological polyanions has been applied on floating thin sections for electron microscopy. The method is based on the formation of insoluble polyanions-quaternary ammonium salt complexes and the subsequent formation of electron dense ion association complexes following a ferric thiocyanate treatment. Mast cell granules and eosinophilic granule crystalloids were selectively stained. Nucleic acid contrast was selective to some extends.
