Abstract

A luminescent bacterial reporter assay, the SOS lux test, was developed for rapid detection of environmental genotoxins. The bioassay is based on recombinant plasmid carrying the promoterless lux operon of Photobacterium leiognathi fused with strong SOS inducible promoter. The DNA damage potency of chemicals or physical factors can be monitored by measuring the visible light emitted by growing culture. The sensitivity of the SOS lux test is similar to other bioassays, such as SOS Chromotest, umu test and Ames mutatest. The role of enzymes of DNA damage-repair system in SOS response of cells can be evaluated by using the appropriate mutant cells as a host cells in this test-system. The cell cultures as well as immobilized cells can be used for detection of genotoxins in SOS lux test.
