Abstract

Recombinant adenovirus vectors are still the preferred choice for many types of gene therapy. However, the availability of a process to purify larger quantities of these vectors at high purity and on a consistent basis for screening and small animal experiments remains a challenge. Classic 2X CsCl gradients are labor-intensive and time-consuming. Alternatives to CsCl-gradient purification based on ion-exchange membrane chromatography have been developed, but results are not consistent and the procedures cumbersome. We report the development of the Adenopure LS™ Large Scale Adenovirus Purification Kit. This kit is membrane adsorber-based and produces high-quality adenovirus in as little as two hours.
