Abstract

Publisher Summary The immobilization of pyridoxal 5'-phosphate (PLP) and pyridoxal 5'-phosphate-dependent enzymes on Sepharose is described in this chapter. Immobilized derivatives of PLP (pyridoxal 5'-phosphate) may be useful for the purification by the affinity chromatography of enzymes requiring this coenzyme. The chapter discusses (1) the preparative procedures for three types of Sepharose-bound PLP and their properties and (2) the application of these immobilized PLP for the affinity chromatography of apo-tryptophanase from Escherichia coli and for the immobilization forms of some PLP enzymes consisting of multisubunits, for example apo-tryptophanase, through biospecific binding to the active center of the one subunit. 6-Immobilized PLP (SP-A) is obtained by coupling diazotized p-aminobenzamidohexyl-Sepharose to the 6-position of PLP.
