Abstract

This chapter discusses the qualitative and quantitative virus-yield assay for antibody neutralization of interferons. An antibody neutralization assay for interferon activity is developed that enables the resolution and quantitation of antigenically distinct interferons in a preparation. This method of assay measures the neutralization of antiviral activity by virus yield and is both objective and a highly sensitive means for determining the degree to which an interferon is neutralized. This procedure is outlined using anti-Le interferon serum to neutralize the antiviral activities of Le interferon on three different cell types that differ in their sensitivities to human interferons. The antibody neutralization curves reflect the amount of antibody needed to neutralize the antiviral activity of the interferon on each cell type and can reveal the presence of antigenically distinct interferons in the interferon preparation. This means of assay can also detect neutralizing antibodies with different specificities in the antiserum.
