Abstract
Background: CTCs count with the immunomagnetic-based CellSearch® system (Veridex) has prognostic significance in pts with mBC, however molecular studies on CTCs are limited by the low number of CTCs isolated in a high background of leukocytes. In this study we combined CellSearch® with DEPArray™ (Sylicon Biosystem) technology to develop a protocol for single CTC detection and recovery, and to optimize whole genome amplification (WGA) and sequencing reactions on single-CTC DNA samples to identify mutations in the phosphatidylinositol-4,5-bisphosphate 3-kinase, catalytic subunit α (PIK3CA).
