Abstract

口腔鱗狀上皮癌細胞 ( OSCC ) 有個令人注目的現象就是遷移和轉移。介白素-6 ( IL-6 ) 已被顯示與許多癌症的增生和遷移有關連。細胞間黏附因子 ( ICAM-1 ) 屬於免疫球蛋白超家族的成員，是一誘導型表面糖蛋白，調節細胞與細胞間相互作用。然而，在人類口腔癌細胞中，IL-6是否會藉由細胞間黏附導致癌細胞的遷移以及和ICAM-1表現的關係還尚未被了解。我們發現IL-6會增加人類口腔癌細胞遷移的現象以及ICAM-1的表現量。使用中和抗體抑制IL-6受體的表現，證實IL-6接受器參與人類口腔癌細胞的遷移以及ICAM-1的表達。IL-6接受器的中和抗體、Syk、JNK或AP-1的抑制劑均會減弱IL-6所導致人類口腔癌細胞的遷移以及ICAM-1的表現量，由此可知，IL-6會激活IL-6接受器、Syk、JNK和AP-1的訊息傳遞。而且IL-6會誘導的AP-1活性，其活性也會被IL-6接受器的中和抗體、Syk和JNK的抑制劑所抑制。我們的研究結果顯示，IL-6會藉由IL-6接受器、Syk、JNK和AP-1訊息傳遞來增加ICAM-1表現，進一步提高人類口腔癌細胞遷移的現象。
