Publisher Summary This chapter illustrates the preparation of bovine rod outer segment membranes capable of regenerating visual pigment with added 11- cis -retinol. Bovine rod outer segment membranes (ROSM) isolated by sucrose density gradient centrifugation are unable to regenerate visual pigment with added 11- cis -retinol. The procedure to be described provides an alternative means of isolating ROSM utilizing glycerol and ethylene glycol. It yields an impure preparation with an active nicotinamide adenine dinucleotide (NAD + )-dependent 11- cis -retinol dehydrogenase capable of regenerating visual pigment with added 11- cis -retinol and NAD + . The entire procedure takes at least 14 hr but could be substantially shortened by use of fresh retina. As such it is usually carded out in two successive days. All work is done under dim red light or the dark at 4 °.
