Abstract

This chapter focuses on the separation of oligosaccharides using high-performance anion-exchange (HPAE) chromatography with pulsed amperometric detection. The chapter discusses the results from coupling HPAE chromatography with pulsed amperometric detection (HPAE-PAD) for the chromatography and electrochemical quantification of underivatized oligosaccharides. Within a group of neutral oligosaccharides of approximately the same size and with larger sialylated oligosaccharides (degree of polymerization 14 to 16), the electrochemical response is a reasonably accurate measurement of oligosaccharide concentration. The response factors for phosphorylated oligosaccharides followed the same rules as that of neutral oligosaccharides with an additional decrease in response related to the number of phosphate residues. Within a group of neutral oligosaccharides of approximately the same size and with larger sialylated oligosaccharides (degree of polymerization 14 to 16), the electrochemical response is a reasonably accurate measurement of oligosaccharide concentration.
