Abstract

Different mitogens added to cytogenetic culture directly influence conventional cytogenetics and FISH results. Especially in the case of cancer patients, the presence or absence of mitogens can yield results with distinct un-related clones, which suggest two different diseases. In our laboratory, CLL samples are cultured for 72 h with one culture stimulated with pokeweed mitogen (PWM), phorbol myristate acetate (PMA), and CpG oligodeoxynucleotide (ODN), and a second culture is stimulated with CpG ODN only. In contrast, B-cell lymphomas are cultured for 72 h with CpG ODN only and cultured for 24 h unstimulated, where myeloid samples are cultured 24 h without any mitogens.
