Abstract

Publisher Summary The best yields of interferon are obtained from human leukocyte suspensions with one particular strain of Sendai virus as the inducer. Cultivation, assay and storage of sendai virus are discussed. The inoculum is a Sendai virus preparation recently passed in eggs. It is diluted 10 -4 in sterile phosphate-buffered saline (PBS) without calcium and magnesium, pH 7.4, supplemented with 25μg of neomycin per milliliter. The sendai allantoic fluid can be stored as such for weeks at 4°. The virus in batches of 2000 eggs is prepared and about 10 ml of sendai allantoic fluid is recovered per egg. Most l-liter batches are sterile. If batches slightly contaminated with bacteria are used for the induction of interferon in human leukocyte suspensions, the crude interferon obtained is usually sterile. The crude sendai virus is used for induction of interferon. However, virus partially purified by high-speed centrifugation and resuspended in human agamma serum to a final concentration of 23mg of protein per milliliter is equally active.
