Abstract

CIViC (www.civicdb.org) is a free, open knowledgebase for clinical interpretations of variants in cancer, with no restrictions on data use. CIViC utilizes public curation and expert moderation. Clinical variant annotation in CIViC consists of 6 types: Predictive, Prognostic, Diagnostic, Predisposing, Functional, and the recently added Oncogenic. Literature derived evidence supporting a given annotation is curated into CIViC in the form of Evidence Items (EIDs), which follow a structured model. Multiple EIDs written for a variant can be summarized into a clinical Assertion, which links back to supporting EIDs along with resources such as population frequency databases, to create a general evaluation of variant pathogenicity or clinical actionability.
