Publisher Summary This chapter provides an overview of human interleukin 1. Interleukin 1 (IL- 1), formerly designated “lymphocyte activating factor,” is a cytokine which was strictly defined to be produced by mononuclear phagocytes and to stimulate T lymphocytes. Two fractions of human white blood cells are being used in our laboratories as a source for IL- 1: the whole mononuclear cell (MNC) fraction of peripheral blood consisting of monocytes and lymphocytes, or the adherent cell fraction which is prepared from MNC and consists mostly of monocytes. IL-1 has been also obtained from cultures of two human macrophage cell lines, U93714 and THP-1. The chapter further explains MNC cultures. The chapter further describes purification of human IL-1.
