Abstract

During fetal development, hematopoietic stem cells (HSC) undergo a huge expansion by combining rapid proliferation with self-renewal. In contrast, self-renewing adult HSC are highly quiescent. Determining the molecular signatures underlying these differences is crucial, however, unraveling cell cycle versus self-renewal associated transcription is a major challenge. To address this, we developed an integrated single cell RNA-seq and live cell-cycle index-sorting technique using Hoechst 33342 (DNA) and Pyronin-y (RNA) staining.
