Abstract

Acid-sensing ion channel 3 (ASIC3) expressed in synoviocytes regulates cell responsiveness to decreased pH and inflammation associated with inflammatory arthritis. The purpose of current study was to characterize the role of ASICs in pH-induced changes in intracellular calcium, mitogen activated kinases (MAPK) and cell death in fibroblast-like synoviocytes (FLS), and if the effects of acidic pH are enhanced in the presence of IL-1β. Experiments were performed in cultured FLS from wild-type (WT) and ASIC3-/-, ASIC1-/- mice, and people with rheumatoid arthritis at time of joint replacement.
