Abstract

RTVP-1 (Related to Testes-specific, Vespid, and Pathogenesis protein) is a novel p53 regulated gene identified using our mouse metastatic prostate cancer model systems. It is homologous to human RTVP-1/GLIPR previously identified in human glioblastoma. Recently we demonstrated that RTVP-1 mRNA and protein are progressively down-regulated during mouse and human prostate cancer progression. We further showed that RTVP-1 has tumor suppressor functions (growth suppression and pro-apoptotic activities) and is inactivated by gene methylation in prostate cancer. Adenoviral vector-mediated delivery of mRTVP-1 in an orthotopic mouse model of metastatic prostate cancer suppressed growth and metastasis through direct apoptotic and anti-angiogenic effects. In addition, extensive intra-tumoral CD 8+ T cell and dendritic cell infiltration and systemic anti-tumor CTL activities were observed.
