The aim of this study was to examine if bacterial outer membrane proteins (Omps) increase the antitumor effect of CA9 in mouse RCC model. METHODS The recombinant OmpA from Acinetobacter baumannii (AbOmpA) was used for immunoadjuvants. Male BALB/c mice of 7 week old were injected subcutaneously with 5 X 105 CA9-expressed RENCA cells and the vaccines were administered on day 3, 10, and 17 after tumor injection. We compared the effect of different conditioning regimens of the murine bone marrow-derived DCs pulsed with tumor lysate alone or in combination with AbOmpA. Tumor growth was monitored individually every 3–4 days by measuring three perpendicular dimensions of each tumor and calculating tumor volume. Cytokine productions were measured using ELISA. RESULTS Pulsing with CA9 and AbOmpA elicites production of Th1-promoting interleukin-12 from DCs and enhances the capacity of DCs to stimulate immune responses. Compared with DCs pulsed with RENCA tumor lysate, the DCs pulsed with CA9 and AbOmpA simultaneously produced more powerful immune reactions. In a therapeutic model of murine RCC, DCs pulsed with CA9 and AbOmpA at the same time significantly decreased tumor growth.
