Publisher Summary This chapter describes the basic cannulation technique for mice as well gives a specific example of its usefulness for immunological studies. It explains a procedure for collecting thoracic duct lymph (TDL) from mice to determine lymph flow and lymphocyte output. Since investigations are done with various modifications to the basic technique and have used it for a wide range of experimental purposes. The chapter reviews that these applications have primarily included analysis of lymphocyte recirculation and the obtaining of T lymphocyte populations which have been positively or negatively selected for specific antigens. It also discusses that the technique of thoracic duct cannulation has been primarily used for experiments where depletion of specific antigen-reactive T lymphocytes is required.
