Because expression profile of microphthalmia-associated transcription factor (MITF) correlated well with IFN-α-response status of RCC cell lines, we focused clinical significance of MITF expression in clinical specimens. In this study, we investigate correlation between expression level of MITF and presence and future occurrence of metastasis in patient with RCC. METHODS From 14 RCC cell lines and 65 RCC samples, RNA was extracted and used in this study. After transfection of cDNA of the MITF, alteration of gene expression level and IFN-α-susceptibility of transfectant were analyzed by quantitative PCR and WST-8 assay, respectively. In RCC samples, mRNA of MITF was measured and correlation between MITF expression level and clinico-pathological parameters were statistically analyzed. In patient without metastasis (M0 RCC) at the time of nephrectomy, correlation between MITF expression and future metastasis was analyzed. RESULTS MITF cDNA transfection into IFN-α-resistant RCC cell line resulted in elevation of MITF expression and acquisition of IFN-α-sensitivity. Statistical analysis revealed that low MITF mRNA expression is significantly correlated with presence of metastasis and poor survival of the patient (p<0.0258 and p=0.0065 (Figure), respectively). In addition, low MITF expression seemed to correlate with poor recurrence free survival of patients with M0 RCC. CONCLUSIONS MITF gene transfection provided elevation of IFN-α-sensitivity in RCC cell line, suggesting that this gene is a target molecule for modulation of IFN-α-response.
