Abstract

To rebuild large-scale 3D tissue architectures, we need to construct not only capillary blood vessel but also small-diameter blood vessel within them. We have developed a method to fabricate patient-specific vascular scaffolds from a biocompatible and biodegradable polymer, PLCL (poly(L-lactide-co-ε-caprolactone)). In this paper, we propose a way to seeding smooth muscle cells and fibroblast cells outside the PLCL scaffold using Reeling Fiber System (RFS). In the first phase, hydrogel fiber containing NIH3T3 cells was reeled onto tubular PLCL scaffold and broken down by an alginatelyase. As a result, we succeeded in seeding cells onto scaffold and the cells viability was more than 90%. We believe that this fabrication technique will be applicable in wide areas such as tissue engineering and bio simulators.
