Abstract
The safe and effective gene delivery system is one of the most important issues of gene therapy. Especially when it applied to treatment for nonlethal diseases, viral vectors should be avoided not to induce the immune responses, and sufficient non-viral systems are strongly demanded. Many kinds of cationic lipids and cationic polymers have been studied, however, the problems such as cytotoxicity or instability under the physiological conditions are not completely resolved. Moreover, the mechanisms from DNA uptake to gene expression inside cells and the cell response which may affect the mechanisms still remains unclear.
