Abstract
Obesity is a state of chronic inflammation and also characterized by compromised immunocompetence. Little knowledge is available in cellular mechanism linking chronic inflammation and impaired immunity. We investigated systemic inflammation and antigen-stimulated intracellular cytokine levels in 96 asymptomatic individuals and effects of in vitro recombinant TNF incubation. We hypothesized that obese individuals will exhibit elevated plasma TNF levels contrasted by impaired antigen-stimulated TNF production by monocytes. Body mass index (BMI) was used to identify overall obesity, and DEXA scan was used to determine %trunk and total fat. Plasma TNF levels were determined by immunoassay. Lipopolysaccharide (LPS)-stimulated monocytic intracellular TNF was assessed by flow cytometry.
