Abstract

Background: Valproic Acid (VPA) is a histone deacetylase (HDAC) inhibitor found to prolong survival of rats in hemorrhagic shock when given as pretreatment. In this study we test whether the post-hemorrhage administration of VPA with small volume resuscitation would induce histone hyperacetylation and inflammatory modulation in a lethal swine hemorrhagic shock model. Methods: Anesthetized Yorkshire swine were subjected to lethal hemorrhagic shock using standard iliac vessel injuries with uncontrolled bleed for 15 minutes (average blood loss of 40% Total Blood Volume, TBV).
