Abstract

Publisher Summary The selection and use of genetic mutants to study complex biochemical problems is a well-established technique of enzymology. Selected mutants have been valuable in the analysis of individual reactions in photosynthesis. Photosynthesis mutants have been isolated from Scenedesmus, Chlamydomonas, Euglena, and other species. These mutants have been widely used in the studies of primary photochemical reactions, electron-transport pathways, and photophosphorylation. In spite of the variety of microorganisms used in genetic studies of photosynthesis, very few good examples exist among higher plants. The main reason is the difficulty of handling the large number of individuals required for such work in higher plants. This chapter summarizes higher plant work and outlines the methods used to select and characterize photosynthesis mutants of Zea mays. The chapter also explains the use of higher plant mutants and describes mutation and genetic handling of maize stocks. The chapter also discusses the screening for photosynthesis mutants and initial testing methods for selected mutants.
