Abstract

It is commonly accepted that immune tolerance to self-antigens occurs in the thymus and secondary lymphoid organs. Despite the presence of rich immune microenvironments in non-lymphoid organs, such as skin and gut, it is currently unknown if self-tolerance is actively maintained in these tissues. We developed a novel technique to induce genetic recombination only in regulatory T cells (Tregs) that reside in skin. Cell lineage-tracing revealed that skin Tregs undergo significant attrition and replenishment during their natural life cycle.
