Abstract

발효된 soycurd에서 가장 높은 GABA 함량을 나타낸 Lactobacillus sakei 383은 인공위액(pH 3.0)에서 3시간 경과 후에도 99%가 넘는 높은 생존율을 나타내었으며 0.3% oxgall이 함유된 MRS 배지에서는 88%의 생존율을 나타내었다. 또한 Lactobacillus sakei 383은 6% NaCl 이하로 함유된 배지에서 생육저해가 낮은 것으로 나타났다. Lactobacillus sakei 383은 저해환의 생성 유무를 통해 Listeria monocytogenes, Bacillus cereus, Staphylococcus aureus, Escherichia coli, Salmonella typhi을 포함한 다양한 병원균에 대항하는 항균물질을 생성함을 확인할 수 있었다. Lactobacillus sakei 383에 의해 발효된 soycurd의 발효 과정 중 품질 특성을 측정하였다. 생균수는 발효 36시간 후에 최대치(<TEX>$10^{11}CFU/ml$</TEX>)를 나타낸 후 서서히 감소하였다. Lactobacillus sakei 383에 의한 발효가 진행됨에 따라 soycurd의 산도는 증가하고 pH는 감소하였으며 12시간 이후에는 일정하였다. 수분, 조회분, 조단백, 조지방 및 조섬유의 함량은 각각 <TEX>$94.88{\pm}0.02$</TEX>, <TEX>$0.22{\pm}0.05$</TEX>, <TEX>$2.38{\pm}0.07$</TEX>, <TEX>$1.16{\pm}0.15$</TEX> 및 <TEX>$0.03{\pm}0.01%$</TEX>을 나타내었으며 총당과 환원당의 함량은 비발효 두부 보다 Lactobacillus sakei 383에 의해 발효된 soycurd에서 더 높은 함량을 나타내었다. 필수아미노산과 비필수 아미노산의 함량은 <TEX>$10.45{\pm}1.04mg/100g$</TEX>와 <TEX>$36.86{\pm}2.59mg/100g$</TEX>을 나타내었다. Lactobacillus sakei 383, which showed the highest GABA content in fermented soycurd, survived in artificial gastric fluid (pH 3.0) up to 3 h, and the survival rate was 88%. L. sakei 383 was tolerant to bile juice during incubation in MRS broth with 0.3% oxgall, and the survival rate was 99%. The survival ratio of L. sakei 383 was high in media containing less than 6% NaCl. L. sakei 383 produced an antibacterial substance against various pathogens, including Listeria monocytogenes, Bacillus cereus, Staphylococcus aureus, Escherichia coli, and Salmonella typhi. The quality characteristics of soycurd fermented with L. sakei 383 were measured during the fermentation period. The viable cell number reached a peak (<TEX>$10^{11}CFU/ml$</TEX>) 36 h after fermentation and then slowly decreased. According to the fermentation time of L. sakei 383, the acidity of soycurd increased and the pH decreased until 12 h, and they were maintained thereafter. The moisture, crude ash, crude protein, crude fat, and crude fiber content was 94.88, 0.22, 2.38, 1.16, and 0.03%, respectively. The content of total and reducing sugar was comparatively higher in the soycurd fermented with L. sakei 383 than in nonfermented soycurd. The essential and nonessential amino acid content was 11.2 and 38.65 mg/100 g.
