Abstract

오차드그래스의 최적 조직배양조건을 확립하기 위하여 8가지 품종의 성숙종자로부터 배발생 캘러스 유도 및 캘러스로부터의 식물체 재분화 효율을 조사하였다. 4주 동안 배양한 종자로부터 캘러스 유도율은 Frontier, Roughrider, 93E, Frode, Potomac, Ambassador, Hapsung 2, Amba 순으로 높은 캘러스 유도율을 보였고 식물체로의 재분화율은 Roughrider, Frontier, Potomac, Frode, Amba, Hapsung2, 93E, Ambassador 순으로 나타났다. 전체적으로는 캘러스 유도율이 높은 품종이 식물체 재분화율도 높은 경향을 나타내었다. 본 연구를 통하여 밝혀진 캘러스 유도와 식물체 재분화율이 우수한 품종은 분자육종을 통한 신품종 오차드그래스의 개발에 유용하게 이용될 수 있을 것이다. In order to investigate the effects of genetic variations of orchardgrass in tissue culture response, calli were induced from mature seeds of eight varieties, 'Hapsung 2', '93E', 'Amba', 'Ambassdor', 'Frode', 'Frontier', 'Potomac' and 'Roughrider', and plant regeneration frequency was compared. Significant differences were observed among the varieties in both callus induction and plant regeneration. Callus induction rate of viable seeds varied from 24.3% to 71.7%. Plant regeneration frequency ranged from 76.6% to 29.7%. 'Roughrider' varieties showed higher regenerability with the frequency of 76.6%. These results can be used not only to provide additional improvements in the plant regeneration frequency from transgenic callus, but also useful for molecular breeding of orchardgrass through genetic transformation.
